PCR product & amplicon sequencing
Choose a sequencing strategy around your amplicon length and question. Targeted verification, long-amplicon characterisation and multi-sample profiling need different project designs.
Where this fits your work
- Confirm a single amplified DNA fragment
- Characterise a longer target that needs extended sequence context
- Discuss multiplexed amplicon projects with a defined analysis goal
Build a complete project brief
| Item | What to confirm |
|---|---|
| Product profile | Expected size, number of bands, primer sequences and purification method. |
| Question | State whether you need sequence verification, variant analysis or community profiling. |
| Platform | Discuss Sanger, short-read NGS or long-read sequencing; no single option fits all amplicons. |
| Controls | Identify negative controls, replicates and sample groups where relevant. |
Define what you receive
- Agree sequence or read files, quality summaries and sample naming.
- Specify alignment, variant or community analysis separately.
- Clarify whether PCR, purification and sequencing are each included.

Prepare a project enquiryQuestions before you begin
Should I send an unpurified PCR reaction?
Ask first. Residual primers, enzymes and nonspecific products can affect the selected workflow. Acceptance and any purification charge are project-specific.
Can sequencing remove amplification bias?
No. Sequencing observes the material produced by PCR; primer bias and amplification errors must be considered in the experimental design.
